摘要:目的 研究骨髓间充质干细胞在内毒素诱发急性肺损伤(ALI)兔模型中的保护作用。方法 实验于2007-10/2008-01在唐都医院中心实验室完成。成年健康雄性家兔20只,其中2只用于培养骨髓培养骨髓间充质干细胞(MSCs),余18只随机分为三组:内毒素(LPS)致ALI模型组(LPS组)、生理盐水对照组(NS组)、MSCs治疗组。LPS组、MSCs组利用内毒素气管内滴注法建立家兔急性肺损伤模型,模型建立成功后30min,MSCs组经家兔右侧颈静脉注射方式给予骨髓间充质干细胞,LPS组给与等量生理盐水,NS组按照以上两组造模、治疗方法均给予等量生理盐水处理,所有动物观察48小时后处死,细胞计数板计算右肺灌洗液中的总细胞、中性粒细胞数、BCA法计算总蛋白渗出量。左上肺石蜡包埋HE染色观察病理变化,左下肺称干湿重,计算干湿比。结果 镜下:LPS组肺泡腔和小气道内有大量炎性细胞浸润,明显出血水肿。肺泡间隔明显增厚,MSCs组肺泡壁较薄,只有少量的炎性细胞浸润;统计学上LPS模型组支气管肺泡灌洗液(BALF)中炎性细胞数目、肺组织干湿比以及蛋白渗出量较NS组有统计学意义(P<0.05);MSCs组较NS组P>0.05,无统计学意义。结论 MSCs可能通过在损伤的家兔肺组织中转化为上皮细胞,从而降低支气管肺泡灌洗液中的炎性细胞、蛋白的渗出,对急性肺损伤具有一定的保护作用。
