用how long可以，但你的句子语法有错误。

或
How much time do they spend (in)carrying the messages from one garage to the other?(sb, spend time <in>doing sth.)
表示花费时间可以用上面两个句型，但不可心用cost,它表示花费金钱或付出代价。它的主语通常是物,宾语是人。如：
The book cost me ten yuan.（=I spent ten yuan on the book.)

2. How long do they spend (in) carrying ...?
如果主语用it的话就不能用cost要用take
cost的主语一般是物
而take的主语一般是形式主语it
主语是人的话要用spend

How long do they spend in carrying the messages form one garage to the other?

第二句不可以
