链格孢(Alternaria)属真菌是一种重要的果蔬采后致病菌,受该菌侵染后的果蔬乙烯释放量增加,对该菌是否能自身合成乙烯、乙烯合成途径方面的研究至今尚未见报道。本试验应用气相色谱法分析检测了来自不同果蔬上的Alternaria属菌自身合成乙烯的途径和检测条件,研究了环境条件对该属真菌乙烯合成能力的影响。结果表明,检测Alternaria属真菌合成乙烯的最佳条件为察贝克固体培养基试管内斜面培养、底物为10mM浓度蛋氨酸、乙烯检测最佳时期为104cfu/L分生孢子接种后3天检测;供试13株菌均能合成乙烯,其乙烯合成途径为蛋氨酸—KMBA途径;而各菌种间乙烯合成能力存在差异,其中最高达1.346μl/h,最低为0.1901μl/h。真菌的乙烯合成能力会受到不同环境条件的影响,低温、黑暗、紫外等能降低乙烯合成,而高温、酸性(pH=?)条件有利于Alternaria属菌乙烯的合成。本研究首次报道了Alternaria属真菌合成乙烯的广谱性,初步明确了它们合成乙烯的途径,为进一步研究乙烯在Alternaria属真菌致病过程中的作用机理,奠定了前期研究基础。
