海洋酵母菌SOD分离纯化的初步研究

摘 要:以海洋酵母菌为材料,采用甲苯法、乙醇-氯仿法、异丙醇法和超声波法对其进行破壁;分离出SOD粗酶液,采用邻苯三酚自氧化法,测定不同破壁方法下SOD粗酶液酶活,选取酶活最高的甲苯法;将菌体用甲苯法破壁得到的粗酶液调节pH除杂蛋白,丙酮二次沉淀后,得到纯化的SOD,其比活为1219 U/mg,收率为5.6%;在25℃~95℃间对SOD进行耐热性实验,结果表明低于55℃时SOD活性基本稳定,65℃时仍能保持79.64%的酶活;纯化后SOD经CHCl3-乙醇,H2O2抑制试验,酶类型为Cu、Zn-SOD;在200~400nm 紫外光下测定酶的最大吸收波长,260 nm时出现最大OD值0.931,呈现出波峰,证明酶类型为Cu、Zn-SOD。
