制革废弃物酶法水解工艺研究

摘 要

论文采用酶法对猪皮削匀革屑进行水解工艺研究。实验根据各种酶水解获得胶原蛋白水解液的产率,选择了水解率较高的一种国产酶和一种进口酶,分别进行正交实验,测定水解率,并对胶原蛋白水解液进行特性粘度测定,得到两种酶对于水解率的最佳工艺,并进行优化实验,同时根据实验结果进行相关单因素实验,分析各个因素对水解率及特性粘度的影响。
实验结果表明,对两种酶来说温度对结果的影响均为最大,水解时间对SG酶的影响次之,对胰酶的影响最小。酶的用量过多或过少都会抑制胶原蛋白的水解,只有在适量的时候内才能使水解率达到较大值。胰酶对胶原蛋白的水解比较强烈,水解程度较高,胰酶水解时用量为0.2%(o.w.f),温度48℃,水解48h,产率达99.0%,水解液特性粘度为37.86pa•s。而诺维信酶SG的作用比较温和,水解缓慢,酶的用量为0.6%(o.w.f),温度35℃,水解48h,产率为51.0%,水解液特性粘度为126.48 pa•s。
