【摘要】目的:了解作业工人外周血淋巴细胞p53及DNA损伤修复相关基因mRNA表达水平与苯暴露环境关系,探讨特异性、敏感性、稳定性好,标本易采集等优点检测指标,应用于苯职业人群健康监护。方法:应用SYBR GreenI嵌合荧光法实时荧光定量RT-PCR分析技术,检测72名作业工人(根据岗位不同分为A组、B组)和29名对照人员外周血淋巴细胞p53及相关基因mRNA表达水平。应用差异表达分析软件REST 2005,比较对照组与作业组间表达倍数差异;同时进行外周血白细胞、血红蛋白和血小板计数;抽检原料甲苯、喷涂剂中苯含量;车间空气三苯浓度监测。结果:A组和B组p53、p21、ku80、Ape1和Mdm2表达水平与对照组比较差异无显著性,A组的Rad51、Bcl-2、Bax、Xpa和Xpc表达水平下调、B组Rad51表达下调与对照组比较有统计学意义。A组的白细胞、血红蛋白和血小板低于对照组,B组血小板低于对照组差异有显著性。结论:苯作业工人血细胞中部分与DNA损伤修复相关基因的mRNA表达水平与非接苯对照组比较存在差异。SYBR GreenI嵌合荧光实时RT-PCR法与REST 2005统计软件,检测和分析基因mRNA差异表达水平方法,适用于职业人群的分子流行病学研究及健康监护。
