丹江口水利枢纽工程位于汉江与丹江汇合处下游800米处，是20世纪60年代中国最壮观的水利工程，也是汉江上最大的水利枢纽工程，由挡水建筑物、发电建筑物、通航建筑物、引水建筑物等组成。挡水建筑物丹江大坝总长2.5公里，工程最大坝高97米，坝顶高程162米，内装6台发电机组，装机容量90万千瓦，平均发电量38.3亿度。

该工程控制流域面积95200平方公里，占全流域面积的60%，具有防洪、发电、灌溉、航运、养殖等综合效益。丹江口水枢纽自建成以来，累计拦蓄洪峰流量1000立方米/秒以上洪水67次，减少淹没下游耕地541万亩，匡算防洪效益262.4亿元，保障了汉江中下游的防洪安全。

丹江口水利枢纽不仅是根治汉江、开发汉江的关键工程，而且也是南水北调中线最佳的水源工程。2005年9月26日，南水北调中线水源工程—丹江口水利枢纽大坝加高工程正式开工建设。丹江口水库作为中线调水工程源头按后期规模完建，坝顶高程将增加到175米，正常蓄水位从157米提高至170米，可相应增加库容116亿立方米，近期年均调水95亿立方米，后期调水可达130亿立方米。
