Sliver staining protocol for proteins

Step Solution Time

Fixation Ethanoc (30%) 75ml Acetic acid glacial (7%) 17.5ml
make up to 250ml with distilled water 30min

Sensitizing Ethanol 75ml Glutardialdehyde(25% w/v)* 1.25ml
Sodium thiosulphate(5%w/v) 10ml
Sodium acetate 17g
make up to 250ml with distilled water 30min

Washing Distilled water 3x5min

Silver reaction Silver nitrate solution (2.5%w/v) 25ml Formaldehyde(37%w/v)* 0.1ml
made up to 250ml with distilled water 20min

Washing Distilled water 2x1min

Developing Sodium carbonate 6.25g Formaldehyde (37%w/v) 0.05ml
