摘要

以新鲜山药为原料,研究山药中多酚氧化酶(PPO)的最适pH值、最适温度、最适底物浓度及添加剂的影响,设计了不同护色方案,以确立山药的最佳护色条件。采用分光光度法研究pH值、温度对酶活性的影响;比较不同护色液对酶活力的影响;结果表明,以邻苯二酚为底物,该酶最适作用pH 为5.8,最适作用温度为20℃,Km=1.82mol/L,Vmax=1.11(Δ OD/min)。本文同时考察了抑制剂对山药多酚氧化酶活性的影响,从而制定出最佳护色措施。
