The patient is sick because there is a 250 bp gel band presents, which indicates the DNA of Mycobacterium tuberculosis.
A control sample is missing in this test. Thus, we are not able to compare this potential patient's DNA sample with a healthy human DNA sample (a control), in order to confirm the pressence of disease in patient.
One of the major problem with PCR is contamination during experiment. This is because PCR is a sensitive technique and it requires extensive care in order to obtain accurate result.
I don't really sure about how to answer the last queation. However, I think tha answer should be CAN, because the effect of drug or treatment may cause the DNA fragment to change in its length, and consequently the gel band will shift to another location (no longer on the 250bp location).
