Dear netters,
I am trying to ligate 719bp fragments with two different adapters(3'ddNTP blocking and 5'phosphorylation).The fragments are 3'-A cohesive end and adapters are 3'T cohesive end, which can avoid adapters or fragment connection in tandom.Then I use the adapters' primer to amplify, but no good result obteined. I suppose adapter-ligation is not successful.
Does anyone have any experience like that ?Any coments and advice would be appreciated.

seems that u've made a tiny little mistake,so what does "tandom" mean?
okay,that doesn't matter in understandin' what u r actually after,point is,i am major in pharmacy and such PCR-experiment ain't involved in our studyin',so guess i'm not goin' to figure out the essentials of your little "trick",nor would i offer u any practical suggestions~~~~sorry~~
