多数肿瘤目前尚未发现其特异性抗原成分或与其直接相关的TAA,细胞性肿瘤抗原易于获取和制备,因而给予肿瘤全部细胞性抗原,由DC去实现对肿瘤抗原的摄取、加工和递呈,简便而有效。
用完整的肿瘤提取物、肿瘤细胞裂解物或肿瘤的总RNA对DC进行修饰,可达到刺激机体抗肿瘤免疫的效果,且对弱免疫原性肿瘤同样适用。
肿瘤全细胞抗原冲击DC致敏T细胞引起免疫应答的机制有肿瘤抗原交叉递呈、旁观者效应、DC与肿瘤细胞形成融合体等。
经比较,用照射法制备的肿瘤细胞致敏DC可产生最有效的抗肿瘤免疫。
这种方法的不足之处在于:制备的新鲜肿瘤细胞纯度不够,肿瘤全细胞抗原中包含自我抗原被DC递呈后可引起自身免疫反应。

At present, elements of specific antigen or the related tumor-associated antigen (TAA) has yet to be discovered in most tumors; cell tumor antigens can be easily obtained and prepared, so it is simple and effective to provide tumor with whole cell antigens, and to uptake tumor antigens, processing and presenting through Dendritic cells (DC).
By modifying DC with complete tumor extracts, tumor cell lysates or total RNA of tumor, the stimulation of body anti-tumor immunity can be achieved, it is also applicable to the poor immunogenic tumors.
The various mechanisms of immune response generated by DC sensitized T cells pulsed with tumor whole cell antigens are cross-presenting of tumor antigens, bystander effect and fusion bodies formed by DC and tumor cells.
After going through comparisons, it is observed that the tumor cell sensitized DC prepared by irradiation can produce the most effective anti-tumor immunity.
