[摇瓶阶段灵芝细胞生长与灵芝多糖产生的研究

本论文通过单因素实验，在摇瓶培养阶段考察了碳源、氮源、pH、发酵温度、培养时间、摇床转速对灵芝细胞生长及灵芝多糖产生的影响，采用硫酸-苯酚法测定不同因素下灵芝多糖的含量。通过选定不同因子来培养，分析并得出摇瓶阶段优化培养条件如下：K+ 、Mg2+ 、P5+ 、VB是菌丝体生长所必需的营养成分。通过前人参考资料得知，最适细胞生长的优化培养基为碳源为葡萄糖3.6%，氮源为玉米浆0.2%，pH值 6.5、KH2PO4 0.1%、MgSO 4•7H2O 0.05%、Vit B10.005%；细胞生长与多糖产量之间相关性的结论：就不同C源对菌种产多糖量的影响而言以葡萄糖为最适碳源，由优化培养基可知其最适宜浓度为4%；就不同N源对菌种产多糖量的影响而言以玉米浆为最适N源，由优化培养基可知其最适宜浓度为0.5%；就不同C源、N源比对菌种产多糖量的影响而言以C/N=1:6为最适C源、N源比；就不同PH对菌种产多糖量的影响而言以pH=5.5为最适pH；就不同时间对菌种产多糖量的影响而言以t=7天为最适时间；就不同温度对菌种产多糖量的影响较而言以T=28℃为最适温度；就不同转速对菌种产多糖量的影响：而言以r=150r/min为最适转速[1]。
