摘要
研究了高效液相色谱(HPLC)典型抗生素(喹诺酮和磺胺混标)的检测方法及最优的实验条件。结果表明:结果表明色谱柱为岛津SHIM-PACK VP-ODS 4.6 mm,150×4.6 mm的反相色谱柱,流动相为0.34%磷酸:乙腈=19 : 81(V/V,pH=2.5),流速为0.45 mL/min,检测波长为280nm的色谱条件下得到分离效果较好的5个特征峰。根据上述色谱条件得到线性良好的标准曲线,其回归方程及相关系数r分别为:Y=2.336663*10-3+b,r=0.9998131;Y=6.361123*10-3+b,r=0.9964310;Y=7.0225*10-3+b,r=0.9964461;Y=7.1415*10-3+b,r=0.9966867。建立标准曲线,进而对(喹诺酮和磺胺混标)进行更进一步的检测,探索最优的实验条件。对实验条件进行讨论,柱温>30℃,特征峰开始出现重峰现象,25℃为最优;缓冲溶液(流动相B)pH>3,峰型发生变化,响应值变高,柱温是控制分离选择性的一个重要变量。

Abstract
Studied high-performance liquid chromatography (HPLC) typical antibiotics (quinolones and sulfa mixed standard) test methods and optimal experimental conditions.
